Modulation of anoikis resistance in MG63 osteosarcoma cells by sclareol via inhibiting Ezrin/Fak expression

چکیده
Osteosarcoma is one primary bone tumor with high risk of blood borne metastasis, thus causing aggravation and patient death. The study of molecular mechanism underlying metastasis of osteosarcoma thus may help to relieve the threat of osteosarcoma. Sclareol has been found to have anti-tumor effects in leukemia, colorectal cancer and osteosarcoma, but with unclear functional mechanism so far. Therefore we treated MG63 osteosarcoma cells with sclareol, to observe the condition of anoikis in cells, in an attempt to investigate the modulation of osteosarcoma cell anoikis by sclareol. MG63 cells were cultured by poly HEMA approach. CCK8 reagent was used to detect the cytotoxicity of sclareol on suspended MG63 cells, in order to determine the optimal dosage of sclareol. MG63 cells were further divided into DMSO and sclareol group, in which real-time florescent quantitative PCR and Western blotting were used to detect the expression of Ezrin and FAK genes. Using RNA interference, intracellular expression of these two genes was suppressed. Moreover, flow cytometry was used to detect the cell anoikis. Compared to DMSO group, sclareol-treated cells had lowered expression of Ezrin and FAK genes, with the mRNA level decreased by 45% and 42%, respectively. The ratio of cell apoptosis was significantly elevated (P<0.05). RNA interference assay showed the enhancement of cell apoptosis by inhibiting Ezrin or FAK gene expression. Sclareol may promote the anoikis of MG63 osteosarcoma cells by inhibiting Ezrin and FAK gene expression.

Sclareol, a plant diterpene, exhibits potent antiproliferative effects via the induction of apoptosis and mitochondrial membrane potential loss in osteosarcoma cancer cells.
The objective of the current study was to evaluate the antiproliferative activity of sclareol against MG63 osteosarcoma cells. A 3‑(4,5‑dimethylthiazol‑2‑yl)‑2,5‑diphenyltetrazolium bromide assay was used to evaluate the cell viability of cells following treatment with sclareol. The extent of cell death induced by sclareol was evaluated using a lactate dehydrogenase (LDH) assay. The effect of s...

The correlation between PAK5 (P21-activated kinase 5) and Ezrin gene expression and chemotherapy resistance of osteosarcoma patients was investigated. The cisplatin (CDDP)-resistance model of osteosarcoma cells SOSP-9607/CDDP was established to detect the cell growth curve. Methyl thiazolyl tetrazolium (MTT) assay was used to detect the drug resistance of cells to chemotherapy drugs. Transwell ...

Osteosarcoma (OS) is the most common cancer of the bone. Chemotherapy is commonly used for the clinical treatment of OS. However, chemoresistance to cisplatin [also known as diamminedichloridoplatinum (II) (DDP)] is a major obstacle for OS therapy, the underlying mechanism of which is not fully understood. The present study aimed to investigate the role of microRNA (miR)-199a-5p in the regulati...
متن کاملATF4 regulated by MYC has an important function in anoikis resistance in human osteosarcoma cells
Anoikis resistance is a crucial step in the process of tumor metastasis. This step determines whether the tumor cells will survive when they become detached from the extracellular matrix. However, the specific mechanism of tumor cells to bypass anoikis and become resistant remains to be elucidated. The present study aimed to determine the internal mechanism of bypassing anoikis through comparis...
متن کاملWnt/β-catenin signaling pathway activation reverses gemcitabine resistance by attenuating Beclin1-mediated autophagy in the MG63 human osteosarcoma cell line
Anaberrant Wnt/β-catenin signaling pathway is frequently implicated in tumorigenesis. However, whether the Wnt/β‑catenin pathway plays a role in resistance to antitumor chemotherapy drugs remains unknown. In the present study, the process of autophagy was assessed following overexpression of the autophagy‑associated gene Beclin 1 in gemcitabine‑induced MG63 human osteosarcoma cells. Autophagy‑a...
